چکیده

Recently, xylanases have expanded their use in many processing industries, such as pulp and paper, food and textile. The production of xylanase by the fungus Penicillium sclerotiorum under submerse cultivation was investigated. Oat spelts xylan and wheat bran were the best inducers of xylanase activity. Optimal xylanase production was obtained in liquid Vogel medium, pH 6.5, at 30oC, under stationary condition during 5 days. The temperature for optimum activity was 50oC and optimum pH 4.5. The enzyme was stable at 40oC, with a half-life of 72 min. and when it was incubated at 45 and 50oC, the half-life was 8 min and shorter than 4 min, respectively. High pH stability was verified from pH 3.5 to 7.5. This enzyme showed interesting characteristics for some process, such as pulp and paper industry, because P. sclerotiorum produces low cellulase level, as well as in food industries.

Xylanase production from Penicillium citrinum isolate HZN13 using response surface methodology and characterization of immobilized xylanase on glutaraldehyde-activated calcium-alginate beads

The present study reports the production of high-level cellulase-free xylanase from Penicillium citrinum isolate HZN13. The variability in xylanase titers was assessed under both solid-state (SSF) and submerged (SmF) fermentation. SSF was initially optimized with different agro-waste residues, among them sweet sorghum bagasse was found to be the best substrate that favored maximum xylanase prod...

Production, Purification, and Characterization of a Major Penicillium glabrum Xylanase Using Brewer's Spent Grain as Substrate

In recent decades, xylanases have been used in many processing industries. This study describes the xylanase production by Penicillium glabrum using brewer's spent grain as substrate. Additionally, this is the first work that reports the purification and characterization of a xylanase using this agroindustrial waste. Optimal production was obtained when P. glabrum was grown in liquid medium in ...

COMPARISON BETWEEN CONTINUOUS AND BATCH PROCESSING TO PRODUCE XYLANASE BY Penicillium canescens 10-10c

Penicillium canescens 10-10c strain was cultivated on barley straw hydrolysate as a soluble nutrient source and as inducer for xylanase production. Barley straw hydrolysate was obtained by treatment of barley straw with NaOH or hot water. In shake flask cultures, NaOH treatment was found to increase the biomass production, but was not accompanied by an increase in xylanase production. The best ...

Optimization of Xylanase Production from Penicillium sp.WX-Z1 by a Two-Step Statistical Strategy: Plackett-Burman and Box-Behnken Experimental Design

The objective of the study was to optimize the nutrition sources in a culture medium for the production of xylanase from Penicillium sp.WX-Z1 using Plackett-Burman design and Box-Behnken design. The Plackett-Burman multifactorial design was first employed to screen the important nutrient sources in the medium for xylanase production by Penicillium sp.WX-Z1 and subsequent use of the response sur...

Purification, characterization, gene cloning and expression of GH-10 xylanase (Penicillium citrinum isolate HZN13)

An extracellular thermostable xylanase (Xyl-IIb) produced by Penicillium citrinum isolate HZN13 was purified to homogeneity using DEAE-Sepharose, Sephadex G-100 and Bio-Gel P-60 chromatography with specific activity of 6272.7 U/mg and 19.6-fold purification. The purification revealed the occurrence of multiple forms of xylanases (Xyl-I, Xyl-IIa, Xyl-IIb and Xyl-III). The molecular mass of highl...
